Bio Laboratory

The biology laboratory is a 40 m2 facility which serves to prepare biological and soft matter samples for measurements at the neutron scattering instruments. It is equipped to handle samples in inert gas atmosphere within a glovebox and pre-characterise samples by UV-VIS spectroscopy and dynamic light scattering. A low-temperature freezer can be used to store samples at temperatures down to -80°C.

Operation:

  1. Push the Button on the right of the display to turn on the device.
  2. Wait untill 18, 2 MΩ.cm is displayed..
  3. In case this value cannot be reached, check the connection of the feedwater or drain some water.
  4. Push the green Button on the right sidebar of the device and turn it clockwise to regulate the outflow rate and drain water.
  5. Push the green button again to stop the outflow.
  6. Push the button on the right of the display again to turn off the device.

Please switch off the device if it is not used for a while to disburden the filters of the ultra-pure water system.

Inside the sample preparation lab, which is located in the neutron guide hall, you can also find a ultra-pure water system.

Description:

The workspace inside the compartment dryer is made of stainless steel. It is readily cleanable and equipped with a saved internal 4-side-round heater as well as a grating of stainless steel. The compartment gets sealed by a fully isolated door (stainless steel) with a double-lock and 4-point-justage as well as a fully galvanized backplane.
The target temperature is reached through heating and venting, so the temperature inside the compartment dryer cannot go beneath the surrounding temperature. In case of a sensor failure the heater is deactivated ca. 10°C above the target temperature. In case of a circuit element failure a secondary relay takes control of the heating procedure ca. 10°C above target temperature. In case of a sensor failure (wrong measured values) or total failure of the regulator the heater will be deactivated through a temperature limiter at ca. 10°C above the maximum temperature of 220°C.

Operation:

  1. Turn the device on-/off by pushing the (on/off) lock knob.
  2. Keep (Set) button pushed to set the target temperature by winding the lock knob.
  3. Ventilation adjustment with the stepless regulator.

Instrument Description:

Thermo Scientific NanoDrop™ 2000c Spectrophotometer measures 0.5- 2 µl samples with high accuracy and reproducibility. The NanoDrop 2000c model offers the convenience of both the NanoDrop patented sample retention technology and a traditional cuvette for sample measurements.
The sample retention system employs surface tension to hold the sample in place between two optical fibers, while the beam path can be adjusted by the instrument (0.05 to 1 mm) and gets normalized to 10 mm. This enables the measurement of very highly concentrated samples without the need for dilutions. Using this technology, the full spectrum (190 – 840 nm) NanoDrop 2000c Spectrophotometer has the capability to measure sample concentration up to 200 times more concentrated than samples measured using the standard cuvette.

Instrument Description:

The Zetasizer Nano S is the system of choice where the highest sensitivity and widest size range is required. It is a particle size analyzer for the enhanced detection of aggregates, measurement of small or dilute samples, or samples at high concentration, plus a molecular weight analyzer for molecular size and molecular weight measurement. Static light scattering measurements enable determination of the second virial coefficient for macromolecule solubility tests. The Instrument incorporates 2 techniques in a single compact unit, and has a range of options and accessories to optimize and simplify the measurement of different sample types.

Dynamic Light Scattering is used to measure particle size and molecule size. This technique measures the diffusion of particles moving under Brownian motion, and converts this to size and a size distribution using the Stokes-Einstein relationship. Non-Invasive Back Scatter technology (NIBS) is incorporated to give the highest sensitivity simultaneously with the highest dynamic size and concentration range.

Static Light Scattering is used to determine the molecular weight of proteins and polymers. In this technique, the scattering intensity of a number of concentrations of the sample is measured, and used to construct a Debye plot. From this the average molecular weight and second virial coefficient can be calculated.
This measurement is very demanding on the sensitivity and stability of the whole system, and means that every element of the design has been optimized to ensure accuracy and repeatability

Short-Instructions: calibration

  1. Rinse the electrode with deionized water.
  2. Place the electrode in a calibration buffer/standard and press CAL.
  3. The meter endpoints according to the preselected endpoint mode after the signal has stabilized.
  4. Run the calibration as described above for the other calibrations buffers (up to 5). Be sure to rinse the electrode with deionized water before placing it into another calibration buffer.
  5. Press End to end the calibration procedure. Alternatively, the meter ends the calibration automatically when 5 calibrations are performed.
  6. Press Save to keep the calibration and continue with your measurement.
